IgE NC: Reagent to Measure Allergen-specific IgE

TEST PRINCIPLE

EIA sandwich method: The target allergen, covalently coupled to IgE NC, reacts with the specific IgE in the patient's serum. After washing away non-specific IgE, peroxidase-labeled antibodies against IgE are added to form a complex.

TESTING PROCEDURES

  • Mix serum and a biotinylated allergen (first reaction).
    The biotinylated allergen reacts to serum-specific IgE (1st complex).
  • A series of reactions occur on the reaction filter (porous glass filter, which is solid-surfaced, immobilized anti-biotin antibody).
    Place blocking solution on the reaction filter to promote a uniform response and to prevent a non-specific reaction (adsorption complexes of non-target allergen).
  • Place 1st complex (see 1.) on reaction filter (second reaction).
    Anti-biotin antibody on a porous glass filter traps the 1st complex.
  • Place peroxidase-labeled anti-IgE antibody on reaction filter and incubate (third reaction).
    A sandwich immune-complex forms comprising an anti-biotin antibody sandwich, specific allergens and peroxidase-labeled antibodies.
  • Using a washing solution, wash away unreacted allergens and labeled antibody (B/F separation).
  • Dispense 3,3',5,5'-tetramethylbenzidine (TMB) solution on reaction filter (peroxidase activity causes coloring).
  • Prepare a calibration curve using standard solutions (calibration).
